Multilocus phylogenetic analysis of Phytophthora infestans isolates infecting potato and tomato in India

Severe incidence of late blight on tomato and potato were observed since 2008 in India. A total of 81 Phytophthora infestans isolates collected from tomato (33) and potato (48) in different states of India during July-to-December cropping season of 2009 to 2014 were analyzed for genetic structure. Multi locus phylogenetic analysis involving six genes of ITS, COX2+spacer region, beta-tubulin, elongation factor-1 alpha, enolase and Heat shock protein-90 were performed to understand the structure of P. infestans populations in relation to host and geographical location. A total number of 30 polymorphic sites were identified in the concatenated data set of all the sequences of length 6002-6005 bases enabled to identify 75 different multilocus haplotypes. Phylogenetic analysis of all the isolates based on individual genes and multilocus dataset couldn’t reveal any variation among the isolates based on host or geographical location. The lack of intra species variation and no significant difference between the isolates based on host corresponds to the presence of single clonal lineage of P. infestans, 13_A2 genotype. This 13_A2 multi locus genotype of P. infestans is responsible for severe outbreak of late blight epidemics on potato and tomato across the different regions of India.Downloads
