An efficient technique for isolation and mass multiplication of Magnaporthe oryzae from blast infected samples

A simple and efficient method has been devised for isolation and single spore culture of Magnaporthe oryzae infecting rice and other graminaceous hosts. Fresh or old blast lesions are incubated in moist chamber set-up for mycelial activation and with spore-drop technique fungal growth observed as Lesion Print (LP) on host extract medium. Through serial dilution plating of macerated colony, monoconidial culture was obtained on rice straw extract agar. For mass multiplication of M. oryzae a combination of rice straw extract and oat meal agar medium enabled quick and profuse sporulation within a short period of 5-7 days at temperature of 25±1°C.Downloads
