Protoplasmic fusion: A biotechnological tool for the enhancement of bio-efficacy of native isolates of Trichoderma spp.

Abstract

In this study, it was intended to incorporate the antagonistic potential of a native isolate, Tr 9 (T. erinaceum) and fast growth of another native isolate, Tr 43 (T. asperellum) in one strain. Protoplasts were isolated using lytic enzyme, glucanex and were further fused using Polyethylene glycol (PEG). Preliminary screening of all the fusants with the most virulent pathogen, Sclerotium rolfsii yielded in the selection of 5 fusants which were then evaluated against 5 important soil borne fungal pathogens viz., Pythium aphanidermatum, Phytophthora capsici, Rhizoctonia solani, Sclerotium rolfsii and Fusarium oxysporum and were compared with that of the parental cultures of Trichoderma  f.sp. cubense. The results revealed that the fusant, F2 exhibited >50 % inhibition on the growth of all the fungal pathogens. It showed cent per cent inhibition on the growth of P. aphanidermatum, P. capsici and F. oxysporum, and a per cent inhibition of 51 and 67 on the growth of R. solani and S. rolfsii which were higher than that of their parental isolates.
