Abstract

F3 segregating population of rice, derived from IRBB13 x Poornima, was inoculated with a bacterial leaf blight (BLB) pathogen; Xanthomonas oryzae pv. Oryzae. Phenotypic screening of the progenies (F4population) was performed to select BLB resistant lines. Previously known microsatellite markers were used for the tagging of BLB resistant gene in those lines. Polymorphism and segregation patterns of the markers were studied by cleaved amplified polymorphic sequence analysis (CAPS). Lines no 3 and 33 were found to be resistant to BLB in phenotypic screening. Markers RM23483, RM2344, SR6, E6a and RG136 were polymorphic and co-segregated as in phenotypic screening. The SSR marker RM23495 and RM23496 was used as cleaved amplification polymorphism sequence (CAPS) marker for selective genotyping, but it did not co-segregate as in phenotypic screening.
