Evaluation of culture conditions for the mycelial growth of Lentinula edodes (Berk.) Pegler

Shiitake mushroom (Lentinula edodes) holds global significance for its culinary and medicinal value, particularly due to its health benefits and nutrient profile, including polysaccharides, vitamins, and bioactive compounds. This study investigates the influence of different liquid media, pH levels, carbon and nitrogen sources on the mycelial growth and biomass production of L. edodes . Five liquid media were tested, with potato sucrose broth (PSB) yielding the highest biomass. Varying pH conditions showed optimal mycelial growth at pH 6.0, while alkaline conditions hindered growth. Among carbon sources, sucrose was most effective for mycelial biomass, followed closely by dextrose. For nitrogen sources, peptone significantly enhanced mycelial growth surpassing other organic and inorganic nitrogen options. These findings highlight the importance of substrate optimization in promoting effective growth conditions for L. edodes, providing valuable insights for commercial shiitake mushroom cultivation.
