In vitro effects of Actinomycin D and Cycloheximide on gonadotropin or 17α-20β-dihydroxy-4- pregnen-3-one-induced maturation of Catla catla

Abstract
Actinomycin D and cycloheximide were used as inhibitors to investigate their effect in the oocyte maturation of Catla catla. Full grown immature oocytes incubated with salmon gonadotropin (SG-G100) and 17α-20β-dihydroxy-4- pregnen-3-one (17α-20β-DP) at the concentration of 1 μg/ml induced 91.7 + 1.9 and 93.7 + 2.6 of germinal vesicle breakdown (GVBD) respectively. A significant decline in the frequency of GVBD was observed when the oocytes were incubated with SG-G100 (1 μg/ml) alongwith different concentrations of actinomycin D or cycloheximide. This suggests that both the transcriptional as well as translational inhibitors inhibit the gonadotropin-induced oocyte maturation.A significant inhibition was also observed when the oocytes were incubated in combination with 17α-20β-DP (1 μg/ml) and different concentrations of cycloheximide. The oocyte maturation was not inhibited when the oocytes were
incubated in the presence of 17α-20β- DP (1 μg/ml) and different concentrations of actinomycin D, which suggests that mRNA synthesis is not obligatory for 17α-20β-DP-induced oocyte maturation. It was also found that the inhibition
of oocyte maturation in cycloheximide treated oocytes somehow extends up to 12 h after which the effects gradually declined
