Standardization of latex agglutination test for rapid detection of Aeromonas hydrophila infection

Abstract
Diagnostic techniques for rapid detection of pathogens in commercial aquaculture are essential to combat diseases. Latex agglutination test is a simple serological test and has been standardized for the detection of Aeromonashydrophila infections in freshwater fish. A total of 48 bacterial cultures were isolated and purified from infected Indian major carps showing either ulceration on the body alone or accompanied by dropsy and degeneration of scales. Out of
48 bacterial cultures tested by standardized LAT, agglutination reaction was observed with 27 cultures. All the 27 LAT positive cultures were also simultaneously confirmed as A. hydrophila by biochemical tests. The LAT negative cultures could be identified either as Gram or oxidase negative or Gram-positive cocci/rods. The study revealed a higher sensitivity and specificity
of the test as all the 27 LAT positive cultures were A. hydrophila.
