Enzyme-linked immunosorbent assay and dot immunoassay for detection of Vibrio spp. in tiger shrimp, Peneaus monodon

Abstract
Plate agglutination test, enzyme-linked immunosorbent assay (ELISA) and dot immunoassay (DIA) were standardised and used for detection of Vibrio spp., the causative agents of vibriosis in shrimps. Different bacterial isolates collected from infected shrimps (Penaeus monodon) were tested. Agglutination reaction was noted upto 1:40 dilution of V. parahaemolyticus antiserum. No agglutination was observed with Aeromonas sp. and Pseudomonas sp. Live bacteria followed by formalin killed bacteria, heat-killed bacteria and soluble antigen were found suitable for use as coating antigen in ELISA and DIA. Cross reaction of V. parahaemolyticus antiserum with V. alginolyticus strains and weak reaction with V. anguillarum and V. harveyi were observed with antiserum dilution of 1:1000. However, no cross reaction with Aeromonas sp. and Pseudomonas sp. was noted indicating the specificity of antiserum for detection of Vibrio spp. DIA being more sensitive, quick and economic, has the scope of being used as rapid diagnostic test for detection of vibriosis in shrimps.Downloads
