Differentiation of Karnal bunt resistant and susceptible cultivars of wheat using RAPD

Abstract

RAPD was carried out to determine difference between two widely cultivated Karnal bunt (KB) resistant wheat cultivars viz. HD 29 and DWL 5023 and one susceptible cultivar WL 711 using nine oligonuclocotide decamers viz. OPM-1, OPM-2, OPM-3, OPM-4, OPM-5, OPM-16, OPM-17. OPM-18 and OPM19. Majority of the primers yielded more than six PCR products. Some of the products were specific to WL 711 that indicated absence of such complementary sequences in the genomic DNA of HD 29 and DWL 5023. Similar difference was obtained between the two resistant cultivars also. One PCR product of 1.0 kb size obtained from only resistant cultivars in OPM-16 reaction showed its probable linkage with KB resistance gene in HD 29 and DWL 5023.
