Abstract

In vitro cultures of potato were irradiated with gamma rays in an attempt to induce resistance to late blight, caused by Phytophthora infestans. Two potato cultivars, Kufri Jyoti and Kufri Chandramukhi were cultured in vitro to the 4-5 nodal segmentstage, after which they were irradiated with gamma rays at 20 and 40 Gy. Irradiated plantlets were cultured for 2 weeks before being subcultured for induction of microtubers. Disease resistance was evaluated in vitro using P. infestans culture filtrateand plants categorized as highly resistant or moderately resistant were tested under natural conditions. When plants were irradiated with a dose of 20 Gy, 12 and 8.6% were highly resistant to the culture filtrate and 8 and 30.4% were moderately resistantfor cultivars Kufri Chandramukhi and Kufri Jyoti, respectively. A dose of 40 Gy induced 3 and 2.3 times more resistant plants for cultivars Kufri Chandramukhi and Kufri Jyoti, respectively. Resistant plants in vitro were also resistant under field conditions. All the plants obtained from a single tuber were not consistent in the disease resistance, suggesting a segregation of the mutant character.
