Phenolic composition and antifungal activity of culture filtrate of Leptoxyphium axillatum

Ethyl acetate fractionation of culture filtrate of Leptoxyphium axillatum yielded a semi-solid oily dark brown material (recovery 0.56 g/L of culture filtrate) rich in phenolics. High performance liquid chromatographic (HPLC) analysis of fractionated material indicated presence of 6 phenolic acids, namely, tannic, caffeic, vanillic, ferulic, chlorogenic and cinnamic acids in various amounts. Concentrations of vanillic and chlorogenic acids were maximum constituting 69.7 % of the total phenolics. Low amount of salicylic acid was also noticed. Twenty one fungi belonging to 9 genera were screened by spore germination assay and significant antifungal activity was observed against Ustilago cynodontis at all the concentrations ranging from 500 to 3000 ppm. At 3000 ppm, growth of germ tubes of the fungal spores of six fungi were totally inhibited. Phenolic acids identified in the fractionated material either alone or in various combinations exhibited prominent inhibitory effect on the growth of the germ tubes of the fungi under investigation.
