Detection and chemical characterisation of siderophores of rhizobacterial fluorescent Pseudomonas

Abstract

Siderophore production by 6 fluorescent pseudomonads (P1 isolated from chilli, P2 - cotton, P3-P5 - groundnut, and P6 - soyabean) was investigated and characterised by their chemical nature and binding properties. The organisms were used to inoculate iron-deficient succinate medium and maintained at 30℃ for 48 h. The cell-free culture filtrates were assayed for siderophores by the FeCl3 test, chrome azural S (CAS) assay and CAS agar plate test. The chemical nature of the siderophores, i.e. hydroxamate, catecholate or carboxylate, was determined by a variety of tests. The results showed production of siderophores by all the test organisms in all 3 diagnostic tests. The CAS activity was maximum in the culture filtrate of isolate P2 and least in that of P6. The siderophores of all the isolates gave positive results for hydroxamate; there was no catecholate or carboxylate siderophore. The pyoverdine nature of the hydroxamate siderophores was evident by their absorption maxima between 407 and 413 nm.The siderophores of 3 strong siderophore producer organisms (P1, P2 and P3), when examined for their binding properties with the metal iron, showed distinct colour reactions indicating a bidentate ligand nature of the hydroxamate siderophores.
