DETECTION OF POTATO SPINDLE TUBER VIROID (PSTVd) USING FLUORESCEIN LABELED PROBE

Abstract
A non-radioactive detection protocol based on fluorescein labeled cDNA probe was standardized in dot blot using gene images random prime labeling module. The protocol allows detection of hybrids upon incubation with an antifluorescein alkaline phosphatase conjugate. The signal was discernible upto a dilution of 1:256. To validate the potential of this technique, in-vitro tissue culture plantlets were screened with this probe and its sensitivity was also compared with 32P labeled probe.Downloads
