The present study was conducted on caput epididymis of twelve buffaloes collected from abattoir immediately after slaughtering during four seasons of year viz; spring, summer, autumn and winter. The cryostat sections of 10-12 μm thickness were cut at -200C and were incubated in different substrates for demonstration of phosphatases and oxidoreductases. The study revealed a variable activity of phosphatases (AKPase, G-6-Pase) and oxidoreductases (SDH, LDH, G-6- PD) and diaphorase (NADH, NADPH) in caput epididymis during four seasons of the year. It was concluded that the activity of AKPase, SDH, LDH, NADH- diaphorase and NADPH-diaphorase was maximum during winter season, followed by autumn, spring and summer. The increased activity of these enzymes may be correlated with increased reproductive activity of buffalo bulls during winter season.
