RT-PCR amplification and in-silico analysis of some putative boiling soluble proteins (hydrophilins) related genes from Triticum aestivum.

Boiling soluble proteins encoding genes, although represent only 0.5% of total genome, yet they play an important role in plants. With the advent of molecular and genomic technologies, emphasis is now being placed on understanding the roles of genes in drought stress tolerance using in-silico analysis tools. In the present study, 4 drought responsive Expresses Sequence Tags (ESTs), designated as TADreb1a, TADHN1, TAWCS 120a, and TAWCS 120b were isolated using RT-PCR using specific primers. Signature sequences related to drought stress were also detected in these genes. Using bioinformatic tools like SMART, a few stress related motifs were observed in TADreb1a, TADHN1, and TAWCS 120a. Â BLAST analysis revealed 100% homology TADreb1a, TADHN1 and TAWCS 120a with dehydration-responsive element binding proteins (DREBs), cold-responsive proteins (CORs) and dehydrins (DHNs), respectively. Based on these results, the possible role of TADreb1a, TADHN1, TAWCS 120a, has been discussed.
