Molecular characterization and genotyping studies of Bacillus cereus isolated from buffalo raw milk and milk products

Abstract
Bacillus cereus, a ubiquitous organism is mainly responsible for foodborne illness. As the comprehensive data regarding B. cereus occurrence is lacking, the present study was conducted in order to determine the incidence, virulence gene and antibiogram profiles along with genetic diversity of B. cereus obtained from different milk and milk products. A total of 31 B. cereus were isolated from 320 raw milk and milk product samples showing overall incidence of 9.68% with 8% (12/150), 15% (3/20), 12.1% (4/33), 11.8% (2/17), 10.8% (4/37) and 21.4% (6/28) from raw milk, basundi, burfi, junnu (colostrum milk pudding), kalakhand and khoa samples, respectively by using PCR. None of the ice cream and flavoured milk samples showed B. cereus by PCR. Of the 31 B. cereus isolates, nheA, nheB and nheC genes were found in 10 (32.3%), 12 (38.7%) and 16 (51.6%) of the isolates, respectively where as entFM was detected in 14 (45.2%) isolates. None of the isolates were positive for the hbl complex and cytK genes. The three component Nhe complex (all nheA, nhe B and nheC genes) was detected in 12.9% (4/31) isolates indicating the possibility of non-haemolytic diarrheal food poisoning syndrome. All the isolates were resistant to beta-lactam antibiotics but highly sensitive to gentamicin and tetracyclin. ERIC-PCR was able to differentiate the B. cereus isolates better than REP- PCR in this study.
