Insights of structural and functional luteolytic pattern in Tharparkar cows (Bos indicus) through Doppler ultrasonography

The study aimed to explore real-time variations in blood flow, progesterone (P4) profile, and related structural changes in corpus luteum (CL) of Tharparkar cows following PGF2α administration. Tharparkar cows (n=8), crossed a minimum of 90 days postpartum with well-developed CL in either of the ovaries and were administered PGF2α (Cloprostenol 500 µg) intramuscularly. Ultrasonographic examinations and blood samples were taken at 0, 0.5, 1, 2, 4, 8, 12, 24, 48, and 72 hrs. post-injection. CL diameter, area, and volume showed significant reductions (p <0.05) at 12 hrs post-PGF2α injection, with progressive regression towards estrus. An abrupt rise in blood flow inside the CL peaked at 1hr post-PGF2α injection and then significantly declined (p <0.05), with the lowest values at 72 hrs. Serum P4 levels significantly decreased (p<0.05) from 1hr post-PGF2α injection, indicating functional luteolysis. Initially, the CL diameter remained unchanged for up to 4 hrs following the PGF2α injection followed by a significant size reduction observed at 24, 48, and 72 hrs. The average CL diameter decreased from 20.29±1.19 mm at the time of injection to 11.55±0.30 mm after 72 hrs. Therefore, these results offer valuable insights into the hemodynamic and structural changes occurring during luteolysis in Tharparkar cows.
