Abstract

The present study was carried out with the objective to identify the causative agent(s) involved in the causation of cutaneous abscessation in goat from the semi-arid region of Rajasthan, India by using the conventional bacteriological and PCR protocols targeting 16S ribosomal RNA gene. While doing the survey of goat flocks in the month of January 2025, pus collected from an adult female Sirohi goat having a large sized palpable cutaneous abscess and cultured on BHI agar plate. The amplicons obtained out of universal 16S ribosomal RNA using the chromosomal DNA isolated from the bacterial colonies grown on BHI agar plate were cloned into E.coli based vector and the confirmed recombinant plasmids were subjected for Sanger sequencing. The organism specific genes, recN and galactokinase were also targeted and sequenced after the conventional gene cloning experiments. The obtained nucleotide sequences of both 16S ribosomal RNA and galactokinase genes of the identified bacterial species from Indian goat were analysed using the standard bioinformatics tools. Upon the sequence analysis, it was found that the obtained nucleotide sequences of the bacteria, were having more than 99% identity with 16S ribosomal RNA gene of Streptococcus pluranimalium. Both recN and galactokinase genes were also having 100 % identity with that of S.pluranimalium. In the phylogenetic tree analysis, S.pluranimalium from Indian goat clustered with S. pluranimalium from other host species. It was therefore concluded that in a Sirohi goat from Western India having cutaneous abscess, S.pluranimalium was isolated and characterized at molecular level for the first time.
