In vitro anthelmintic activity of aqueous extracts of Coriandrum sativum seeds and Swertia chirata against Haemonchus contortus in Barbari goats

Abstract

The present study evaluated the in vitro anthelmintic efficacy of aqueous extracts of Coriandrum sativum seeds and the whole plant of Swertia chirata against Haemonchuscontortus isolated from Barbari goats. Strongyle eggs were identified microscopically and cultured to obtain infective third-stage larvae (L3), which were molecularly confirmed as H. contortususing PCR amplification of the ITS-2 region of ribosomal DNA. Anthelmintic activity was assessed using the egg hatch assay (EHA) and larval paralysis assay (LPA) at extract concentrations of 12.5, 25, 50, and 100 mg/ml. Closantel (0.5 mg/ml) and phosphate-buffered saline served as positive and negative controls, respectively. The aqueous extract of C. sativum seeds inhibited egg hatching by 27.49-86.94% and reduced larval motility by 35.07-89.93% in a dose-dependent manner. In comparison, theS. chirata whole-plant extract exhibited significantly higher efficacy, with egg hatch inhibition ranging from 43.64 to 93.12% and larval motility inhibition from 47.40 to 94.10%. Closantel achieved complete inhibition in both assays. The findings indicate that the aqueous extracts of S. chirata and C. sativum, particularly the former, possess considerable in vitro anthelmintic activity against H. contortus and may serve as promising phytotherapeutic alternatives to synthetic anthelmintics, warranting further in vivo evaluation.
