Curcumin and L cysteine mitigates cryopreservation-inducedcryodamage in Barbari buck semen

The study aimed to evaluate effect of different concentration of cucumin and L cysteine HCL in mitigation of cryopreservation-induced cryodamage in Barbari buck semen. The present experiment was carried out on four Barbari bucks, 2-3 years of age. Twenty-four ejaculates (six from each buck) were collected and diluted with Tris-Egg-Yolk-Citric acid-Fructose-Glycerol extender containing no additives (CON; control), Curcumin 1 and 2μg/ml (CUR1 and CUR2), L cysteine HCL, 2 and 5mM/L (LC2 and LC5) at 37 ºC and cryopreserved following standard protocols, processed, frozen, and evaluated for various seminal markers. As compared to control, the motility (50.50±0.42, 53.50±0.50, 52.50±0.50 and 51.33±0.42%), liveability 77.67±0.98, 81.50±1.05, 81.00±0.96 and 77.67±0.88%) and HOS reactivity (39.83±1.10, 44.00±0.96, 43.00±0.51 and 41.17±0.98%) was significantly (P<0.05) improves, whereas abnormality (11.83±1.07, 7.66±0.55, 8.66±0.49 and 11.00±1.26%) and enzyme leakage i.e. GOT (156.5±6.42, 140.0±5.19, 147.8±4.67, 151.8±5.73μmole/L), GPT (243.7±7.67, 231.5±6.22, 234.2±6.21 and 241.8±6.94 μmole/L), ACP (49.50±2.07, 47.33±1.87, 49.33±1.76 and 51.83±1.85KAU/100 ml) and AKP (140.5±5.28, 134.8±4.76, 137.0±5.02 and 145.5±5.98 KAU/100 ml) significantly decline in CUR1, CUR2, LC2 and LC5 groups respectively.In conclusion,supplementation of 2μg/ml Curcumin and 2mM/L seems to be beneficial in mitigating cryopreservation-induced cryodamagein barbari buck semen.
