Effect of biochemical characteristics of buffalo oestrual mucus on in vitro sperm migration

Sperm mucus penetration test was conducted in vitro on 44 oestrual mucus samples collected from Murrah buffaloes during early - (group 1), mid-(group 2) and late-heat (group 3). The mucus samples from animals who subsequently got conceived registered sperm penetration more than 20 mm/20 min. Based on this fact, the samples were categorized into 2 classes (grade 1 and grade 2). Highest conception rate was observed in group 2 where sperm penetration was maximum. The sperm penetration through oestrual mucus depends mainly on its sodium and protein contents since group 2 samples were found to contain significantly (P<0.05) higher sodium level and lower protein contents than others. However, the values for pH, potassium, calcium, magnesium and chlorides did not vary significantly (P>0.05) between 2 classes and among 3 groups.
