Pharmacokinetics of cefazolin in healthy and febrile goats and its interaction with diclofenac sodium

The present study was conducted to study the disposition kinetics of cefazolin (10 mg/kg) in healthy and febrile goats and its interaction with diclofenac sodium. Peak plasma cefaolin concentrations, determined by high-performance liquid chromatography (HPLC), were 12.20 and 2.64 μg/ml in febrile goats following i/v, and i/m administration respectively. Elimination half-life and clearance in healthy goats were 1.18 and 3.36 L/kg/h following i/v administration and 0.97 and 2.26 L/kg/h following i/m administration, respectively, while in febrile goats 1.67 h and 2.59 L/kg/h following i/v administration and 1.08 h and 2.05 L/kg/h following i/m administration, respectively. Concurrent administration of diclofenac increased plasma elimination half-life to 2.98 h; 1.79 h and decreased clearance to 1.48; 1.67 L/kg/h as compared to febrile goats without diclofenac following i/v and i/m administration, respectively. Volume of distribution (Vdarea) and area under curve (AUC) in healthy goats were 4.47 L/kg and 3.00 μg/ml/h following i/v and 3.14 L/kg and 4.46 μg/ml/h following i/m administration, respectively, while in febrile goats were 5.80 L/kg and 4.15 μg/ml/h following i/v and 3.36 L/kg and 5.44 μg/ml/h following i/m administration, respectively. Diclofenac coadministration with ceftriaxone slightly increased the volume of distribution of 6.01 and 4.22/kg and decreased AUC to 2.96 and 1.93 μg/ml following i/v and i/m route of drug administration, respectively. Finding of present investigation indicated a favourable pharmacokinetics of cefazolin in healthy and febrile goats and in combination with diclofenac in febrile goats.
