Modified method for generating microaerophilic environment for isolation of Campylobacter fetus

Abstract

Campylobacter fetus is microaerophilic as it requires 5% O2 10 % CO2, and 85 % N , for isolation and cultivation. A modified method was standardised for isolation of Campylobacter fetus. The folumtion, The 80 preputial washing and 50 semen samples in this study, were processed for isolation of Campylobacter fetus by using this method. Sodium bicarbonate, citric acid and sodium borohydride were incorporated in this modified method of creation of microaerophilic environment. The  15 isolates of C.fetus were isolated from 80 preputial washings. All the biochemical tests for identification of these isolated were done under microaerophilic environment. In the present study. 36 samples were also screened by candle jar method and they showed no growth. When these samples were preocessed under microawerophilic environment created by this modifies method, 7 out of 36 samples revealed growth of C. fetus.
