Embryo sexing with heterochromatin staining technique

The embryos were produced in vitro for the study of their development and sex identification by sex chromosomes (cytogenetical method). The embryos were examined to identify the x- and Y-chromosomes and thus sex of the embryos. Conventionally stained embryos were examined for the location of metaphase plates and identification of sex chromosomes. Subsequently the embryos revealing metaphase plates were C-banded to further increase the sex diagnosis. Among the fixed embryos 936/1251 (74.92%) showed metaphascs. In conventional staining 524/936 (55.98%) embryos could be sexed which upon heterochromatin staining revealed 695/936 (74.26%). Chromosome analysis of conventional staining in conjunction with heterochromatin staining is very accurate technique for sex diagnosis in preimplantation embryos at various stages of development from cleavage to hatching besides confirmation of sex it can also be used for other chromosome anomalies afflicting development, growth reproduction etc. later in life.Downloads
