Restriction enzyme analysis of EDS-76 virus Indian strains*

Abstract

Indian strains of EDS-76 virus, viz. EDSV -IVRI/AD-86, Bangalore strain and Madras strain were used to study the restriction enzyme pattern. Strain 127 was used as the reference strain. The 4 viral strains were propagated in duck embryos and then purified by sucrose cushion ultracentrifugation. The DNA was extracted from the purified virus samples by modified phenol: chloroform method and was then subjected to restriction enzyme analysis using 5 endonucleases, Eco RI, Bam HI, Hind III, Pst I and BgII, which yielded 4, 4, 10, 10 and 6 number of identical restriction fragments with all the 4 strains. EDS-76 virus isolates from different regions of the world were classified into groups 1, 2 and 3 based on the restriction enzyme analysis. Strain 127 is classified under group 1. The 3 Indian strains had the same restriction pattern as that of reference strain 127 and it is concluded that these 3 Indian strains can be classified under group 1 along with strain 127 of EDS-76 virus.
