Genetic polymorphism of BoLA-DRB3.2 locus in crossbred cattle as revealed by PCR-RFLP analysis

Abstract

The present investigation was undertaken to explore DNA polymorphism in bovine class II DRB3.2 locus using polymerase chain reaction (peR) and restriction enzyme (RFLP) analysis. Genomic DNA was isolated from 21 Karan Fries (KF) and 20 Karan Swiss (KS) crossbred animals. A 304 bp fragment of the bovine lymphocyte antigen (BoLA)DRB3 gene-exon 2 was amplified by peR and the amplified product was digested independently with RsiYI, HaeIII and Rsa I enzymes. Digestion analysis revealed 3 BstYI, 6 HaeIII and 15 RsaI restriction patterns. All thesc patterns have previously been reported in cattle. In addition analysis of combined patterns of the 3 enzymcs resulted in 4 probable new alleles, viz. kba, tbb und tbe in KF and dba in KS.
