In vitro capacitation of pre-freeze and post-thaw buffalo cauda epididymal spermatozoa

Buffalo cauda epididymal spermatozoa were harvested from mature bulls slaughtered at the slaughterhouse. In vitro capacitation of the epididymal spermatozoa was done by incubation in BO media supplemented with 10 ~lg/ml heparin for 6 h and was evaluated by induction of acrosome reaction by incubating with the fusogenic lipid lysophosphatidylcholine. Significant increase in acrosome reaction with the increase in incubation period was observed. The acrosome reacted status of the cryopreserved spermatozoa was significantly higher than that of the pre-freeze spermatozoa at all the periods of incubation.
