Purification and characterization of protein(s) from placental extracts in buffalo

Pregnancy associated proteins in buffaloes placental extracts were isolated, purified using affinity chromatography and also characterized by SDS-PAGE and western blot analysis. Purified fractions had lower OD values than the nonpregnant tissue extract (NPTE), placental extrnct (PE) and elute fractions indicating presence of some specific proteins in the purified fractions, which may be associated with pregnancy. SDS-PAGE analysis of purified fractions showed presence of 6 polypeptide band of different molecular weights (78,67,53,42,33 and 26 kDa). Out of6 polypeptides, only 2 polypeptides (67 and 53 kDa) were observed immunoreactive in the western blot analysis. Results indicated presence ofcertain proteins associated with pregnancy in placental extracts and in future its further purification and specific hyper immune serum will be useful to develop specific and sensitive methods for the diagnosis of early pregnancy in buffaloes.Downloads
